I am going to study migration in a549 cell line after a treatment but the a549 cells which I am working with show no migration into the space created by a yellow tip even in normal condition (with no treatment) while cells proliferate in other directions. I think these cells have got some mutations during several passages...am I right or some thing else is wrong?

If your cells has already formed a mono layer then after scratch, it will heal the wound. What you can try is:
1) Thaw new cells
2) Seed them in different concentrations.
3) Its also difficult for over-grown cells to heal the wound because the scratched surface start floating.
4) Use a different cell line.
5) Try a scratch with white tip (10ul) and see if it works or not.

I am really sorry for my delayed response, I' v just noticed your question. I thawed a new batch of cell vial and then my problem solved.
by the way at first I was wrong about the procedure of healing, I expected that healing be faster by covering the gap between scratches with cells rather than measuring areas and comparing them between treated and normal plates.
