You could use active replication as a marker of S-phase. Add EdU or BrdU to the cell culture media wait 30 minutes harvest cells and stain for EdU or BrdU. Big advantage here is that there are some very well characterized antibodies out there for these targets. Of course this would be limited to ICC. For the western I would use Cyclin levels.
