Hi has anyone had experiance of GL-261 not growing subcutaneously? I have inoculated 10e6 cells in C57BL/6J female mice and the tumors became palpable after a week cca but have subsequently disappeared except in few mice. The cell line is routinly used in the literature s.c in imunocompetent strains so I don't know what's the problem.
I frequently used GL261 in commercially available Bl6 mice for either s.c. (with even less cells) or intracranial implantations and never had problems with tumor uptake, or growth. Nevertheless, i ran into problems with mice that were supposed to be Bl6, but seemed not to have had a pure Bl6 background. I suppose you checked the cell viability before the transplalntation, assuming that this was not the problem. To increase tumor uptake and growth in immunocompetent mice (not GL261, but immortalized cells, though), i also used to s.c. inject the cells not re-suspended in pure PBS, but a 1:1 mixture of PBS and Matrigel, which helped to support tumor cell survival and growth after the s.c. injection.
Thank you for your help! I use the same strain of mice for B16 melanoma and EG7 lymphoma cell lines (both derived from Bl6) and never had a problem with tumor growth. Presumably they would also reject these tumors if they were not syngenic. I'll try using Matrigel :)
