Dear colleges! I am currently working with Gibson cloning, however I am pretty new on this. I would like some recommendations : articles, publications, your personal experience which describes in detail the Gibson cloning principle, the advantages and disadvantages. I would like to know your experiences in working with this type of cloning.
Advantage: works almost every time, you can assemble multiple DNA fragments, which saves time, pretty straight forward (PCR, clean-up, assembly, transformation).
Disadvantage: slightly more expensive than classical cloning when it comes to consumables, as you need a lot of different primers that you have to order (which is of course compensated by the fact that it always works and hence saves a lot of time and work).
