I want to dry my plant sample (leaves) by freeze drying.Can someone give me a simple and detailed protocol on how to freeze dry in a lyophiliser . Like how to prepare the sample before freeze drying.Kindly help.
The procedure is so simple. You need to put the plant leaves at -80°C for a couple of hours (ca. 2-4 hours) before applying freeze-drying. Then, put your -80°C freezed leaves on the tray of the lyophilizator in one layer form. Then, start the lyophilizator machine. The drying time can reach 20-24 hours depending on the type of machine.
Is it necessary to keep the plant sample at -80 degree Celsius temps? Because after below zero degree temp the plant sample would be freezed.so why is it necessary to keep the sample at below --80 degree C.
As you know during the freeze-drying process, the sublimation of water occurs at about -50-55 degrees Celsius. lyophilization is a two-step process: freezing then drying. Both steps occur at vacuum conditions (low than 1 mAtm).
So, when you previously freeze your sample at -80 degrees Celsius you improve and accelerate the process. I advise putting the sample holder and tray covered with aluminum foil also at -80 degrees Celcius for the same purpose.
