As a part of my dissertation I am subjecting colonies of E. coli to varying pH levels using hydrochloric acid. The colonies are grown over cell growth quantifiers and six hours into growth, when the exponential phase begins, varying volumes of HCl are introduced using liquid injection systems. The expected result was to see that as the pH decreases, the growth rate would conversely decrease. This did happen but not before there was a massive spike in backscatter that the sensors recieved. This left the OD600 for each of these results peculiar as when the HCl is introduced it indicates a population explosion, followed by instant population decline. Any potential solutions to this would be much appreciated as it has both my lecturers and I very confused.
