I am new to MTT assays and research in general. I am performing a MTT assay of the same cell line with 4 different conditions.
How would you do biological replicates in this situation? I know I have three technical replicates for each condition. However, if I were to do biological replicates, does that mean before I plate the cells in the 96 well plate, I should start with 3 different plates of the same cell? Or are biological replicates in MTT assays considered repeating the same experiment 3 times?

For MTT triplicate is required for data varification for each different conditions. So if you have different sample from different sources, you have to treat them separate and use a triplicate for a reliable and we'll accepted data for scientific evidence.
Alyssa Kim As you mentioned you are using same cell line , if you want to vary the concentration of a molecule you can use the same plate just label it one one side in a coded form to understand the change in concentration to be added.
Same can be done to see effect of substance or drug or hormone. Thus you perform all in one plate and perform accordingly to triplicate the experiment. So you save your time and consumables used for performing in three different plates. Hope it was of help!

Biological replicates means repeating the experiment three times to ensure a 2/3 rd probability that the average results that you obtain is accurate than what you would have obtained by doing the experiment only once.
So you will have to run four experiments using the same cells three times since there are four different conditions.
In addition, you will have to run a triplicate for each condition as you have already mentioned to increase the credibility of your results.
