To ligate blunt insert into blunt vector does the vector need to be dephosphorylated at the time of ligation? I have digested my vector with restriction enzyme that create sticky ends that linearize it. Shall I dephosphorylate this linearized vector before blunting or do the other way around? Is the presence of 5' phosphate necessary in vector to go ligation?
