The antidepressive and anxiolytic efficacy of flupenthixol and flupenthixol decanoate have been investigated in some controlled and open trials in patients with depression. Its antipsychotic effects are likely caused by D2 and/or 5-HT2A antagonism, whereas its antidepressant effects at lower doses may be mediated by D2/D3 autoreceptor blockade, resulting in increased postsynaptic activation. Probably like any other antipsychotic drug it has anxiolytic and anti depressive effect in low doses, specially those with D2 and 5-HT action. Sulpiride, amilsulpride, aripiprazole and quetiapine (and many others) seem to have the same profile and fight for the same indication. Although useful in some cases of depression specially when associated with an antidepressant drug, many studies only seem to search for new indications for low-selling drugs. Extrapyramidal adverse events, weight gain, and increase prolactin serum level might appear during treatment are a disadvantage of the use of antipsychotic medication in depression. But the antipsychotic may be a good option for potencialize the treatment in resistant depression.
