I am trying to express a protein which according to literature is very unstable and toxic. I tried to add some fusion domains to make it stable and express it. Currently I am using C43 DE3 pLysS strain, as it was good for expressing toxic proteins. There is still some degree of toxicity as cells take so long to grow but I was able to purify the protein. However, after running SDS gel I found that there is clearly a lot of expression but the molecular weight is actually lesser than expected. I added the protease inhibitors during purification so I don't think it degraded during purification. At the gene level it is fine according to sequencing results, I believe that is because of the protein's instability and cell's housekeeping proteases. I was wondering if I could get some recommendations about some other strains which have knockout housekeeping proteases or that might be useful in expressing unstable proteins.
