Concerning the DNA extraction, using cow's blood, i suggest you use the Buffy coat, avoiding excessive red cells. Just like 300 microlitres is ok for the extraction.

I suggested that you can sum many low DNA sample and precipitate them using Alcohol 100% (2.5 volume) after overnight incubation at -20C. Use centrifugation at 140000 rpm for 25-30 min.
