Hi~ I need some information about RL95-2 cell.
I'm working on the research that related to endometrial cancer, so I use human endometrial cancer cell, RL95-2. I bought RL95-2 cell from ATCC. ATCC recommends to culture RL95-2 cell in DMEM:F12 with 0.005 mg/ml insulin and fetal bovine serum to a final concentration of 10%. So I'm using DMEM:F12 media with 0.005mg/ml insulin + 10% FBS + 1% penicillin/streptomycin.
But my my RL95-2 cells grow too slow and grow like a colony in a very remote locations. I read some papers that uses RL95-2 cells to perform wound healing assay, so I'm wondering if I'm raising cells in wrong conditions.
Any information about RL95-2 cell would be extremely appreciated.
Complete Growth Medium
The base medium for this cell line is ATCC-formulated DMEM:F12 Medium Catalog No. 30-2006. To make the complete growth medium, add the following components to the base medium:
0.005 mg/ml insulin
fetal bovine serum to a final concentration of 10%
Subculturing
Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 mL of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37°C) until the cells detach.
Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:5 is recommended
Medium Renewal: Every 2 to 3 days
Cryopreservation
culture medium 95%; DMSO, 5%
