1) may i know the best approach to elucidate the possible pathways coming from any membrane bound proteins. currently, the cytoplasmic binding partners of these proteins are not known. I'd probably have to identify the binding partners first before i can even hypothesize about the phenotype governed by these pathways?
Probably, I'd mainly go through a genomic collector web-site like GeneCards. Searching a gene through its interface, you might browse the pathways it is involved in. You might do the same by KEGG and looking for your gene by its searhc engine.
If, instead, its functions or pathways of belonging are unknown, you may want to proceed as you mentioned, and thus look for its partners. You can do it simply by BioGRID, GeneMANIA or STRING and verify their functions or pathways.

If the partners are not known, you might want to use BioTag (Biotynilation of everything that's close to the tag) of the intracellular/extracellular domain and then pull down the proteins using Avidin beads. Then, send the sample for Mass Spec and look for the top hits. People have used this to find partners of membrane receptors.
