I'm working with C57Bl/6J and I have two distinct groups of diet: chow diet and high fat diet. I'm having problems that my animal fed chow diet are with high fasted glycemia, aroung 130-160mg/dl. What could be causing that? Because with these results, it's almost impossible to compare with hight fat diet fed group.

In my opinion C57Bl/6J is a much sensitive strain for metabolic studies. For this hyperglycemic condition there must be the following causing factors:
1. Most imortantly, Composition of the chaw diet you are using (please get a composition ceritificate from the manufecturer or do the analysis by urself)
2. Age of the mice (make sure you are using mice of same age).
3. There should be no external stress (or any kind of) to the animals (stress, sympathatic activation and endocrine alterations could directly influence the blood sugar).
3. Time of sampling and testing method also need to be checked.

Are you obtaining your blood samples from anaesthetized animals by any chance? In our experience anaesthesia can cause high glucose values due to poor breathing accompanied by adrenal discharge.
At what age are you measuring blood glucose? The younger the higher the blood glucose.

Rafael Calixto Bortolin our mice have more than 4 months of age.. and we are using high fat diet 60% of fat.
I had the same problem during my graduate work that came predominantly from handling stress, which can cause a large blood glucose spike. To ameliorate this problem, I used a battery of IP saline injections (100ul every 2 days) leading up to GTT/ITT experiments to acclimatize the animals to handling stress and saw a decrease in fasting glucose from 150-180mg/dl to 100-130 mg/dl.
Also, ditto what Ivan said, anaesthesia will cause a sustained glucose spike as well.
