I am currently in the process of doing my Bsc project and would like people's opinions on the statistics, if possible?

I am looking at a new processing and cutting method for muscle biopsies in electron microscopy and comparing it to the one we currently use. The areas I was going to look at were -

The techincal side - How easy the resin block was to cut on the ultramicrotome etc - Scored 1-5

Semi thin sections - Cell preservation, artefact, contrast etc - Scored 1-5

Ultra thin sections - Are organelles easily recognisable? In terms of contrast, resolution, artefact, etc Scored 1-5

As both new and current processing, cutting methods would have the above areas subjectively scored by a pathologist, what would be the most appropriate test for the null hypothesis - if there is no difference?

I was looking at Cohens Kappa, but wanted to know if others would be more suitable?

Any help would be appreciated!
