I was wondering whether cancer-associated fibroblasts (CAFs), not being cancer cells or immortalised, remain differentiated over several passages in 2D cultures. In the absence of the factors that "educate" normal fibroblasts into CAFs, do they keep their molecular profile? I tried to find literature on this, but all I found were suggestions on the maximum number of passages for studies using CAFs.

I don't have a straight answer to your question, but maybe the easiest way to check it would be to run qPCR for the known expression markers of CAFs, and try to compare results within different passages.
Hi Luisa! I am not on this field but I would search for some particular "phisical" feature of your cells. For example. Are all of the CAF cells presenting EMT? if so, you can focus to the property of mesenchymal cells adhere to plastic. Is the way to "purify" them from an hematopoietic culture. On this sense, I would use this "physical" property here to maintin a CAFs population (but see it as an example of a solution. Probably you can think in another better property). But I would stick to a property that is known to be present on 100% of the cells to be "impartial" on the global population. OR, I would start all the experiments using that kind of property in order to maintain a control on the population of CAFS you are working with. For example, if a mojority of the cells present EMT that example I gave you can be a possible solution.
