I am having difficulties with my project. To induce my Immortalized murine myoblasts H2K A/J (-/-), I've chosen to use EBSS starvation. The problem is that when I incubate the cells for 2hrs in EBSS, massive cell death occurs.

Here are my thoughts:

The EBSS solution I am using does not have adequate nutrients

I am worried that the lack of phosphate may be causing cell death and am tempted to switch to this ebss:

What are your thoughts? I am wondering if it is necessary to have autophagy induction. The non-induced conditions are working and showing up in western blots. Can I not just compare the bafilomycin treated conditions?
