Hello Fellow Scientists,

I'm attempting to detect the amount of LC3-II in A/J and SJL Dysferlin Deficient Immortalized Skeletal Muscle Cells. The issue I am having is that my EBSS condition is causing massive amounts of cell death. The cells are incubated for 2hrs in either EBSS or EBSS containing 100nM Bafilomycin.

I am currently using EBSS Cat No. 14155063 https://www.thermofisher.com/order/catalog/product/14155063

I am fearful that my cells are dying due to the lack of Phosphate and am tempted to switch to this EBSS Cat No. 24010043 https://www.thermofisher.com/order/catalog/product/24010043

What do you think? I am also thinking that complete amino depletion may be to harsh for skeletal muscle cells and may need to supplement with l-Glutamine.
