Hello guys,
I am running western for the 3rd time for the same protein. First two times I can see clear one band. It is inducible protein.
However, on the 3rd time, I have two bands , instead of one band, and no induction (The same two bands in both ctrl and stimulated sample),
the only different thing I made in the 3rd time is that I used less concentration dose to stimulate my gene of interest.
Anyone knows why I see two bands instead of one? anyone experienced this issue before ?

Band splitting can be the result of posttranslational modifications that either change the charge (phosphorylation, Lys-acetylation, ...) or the size (proteolysis, glycosylation, ...).
