It seems that no matter how you align bulbs or set Koehler illumination on a microscope, the image from the camera is plagued by uneven illumination. If you have a scientific camera with appropriate software, you can take a flatfield or blank field image and then the software takes care of correcting for uneven illumination. But how do you solve this problem when you don't have that feature in the camera software, you don't have a flatfield image, or when it becomes a hassle to flatfield correct in downstream software?
