Dear Research Community,
I have used the 35S::RUBY vector from He et al (2020) [Addgene plasmid # 160908] for both agrobacteria and gene gun transformation of Populus x canescens. The transformation efficiency is quite good each time (red dots on the transformed plant material), but there are only a small number of red regenerating plants and they are deformed.
Has anyone also observed similar problems when using the RUBY marker?

I have not worked with the RUBY marker. However, 35S:GFP can cause a same outcome where the plants and/or tissue culture samples with high expression levels of GFP also have poor growth. It may be a similar issue. You could try regeneration of the weaker expression spots in hopes of getting healthier transformants.
