I'm doing pH imaging to investigate pH regultion in cardiomyocytes. To adress the question of the role of NHE, I want to block it selectively. Previously I used EIPA for that, but when I did experiments with the membrane bound pH sensor DHPE fluorescein, I came to know that EIPA is showing fluorescence when excited with 405 nm laser (see also: Gold et al., 2010: "A Clathrin Independent Macropinocytosis-Like Entry Mechanism Used by Bluetongue Virus-1 during Infection of BHK Cells").
So, now I'm thinking to use another inhibitor such as cariporide, eniporide, sabiporide or zoniporide (Avkiran, 2008: "Blowing off acid: a new tool to study Na+/Hco3- co-transport). Does anyone have experience with fluorescence of those inhibitors?
