I am studying the association of certain SNP with a specific disease, and due to several reasons I will use PCR-RFLP method for SNP genotyping, now I have two options:
a- To design and optimize my own PCR-RFLP method .
b- Or use previously optimized method
Which option is good for my research originality?

If the commercially available primers are good enough for elucidating the information you are seeking, I think you should use them (no need to re-invent the wheel :)) unless of this is a methodology development research. The originality of the research comes from your objectives, outcome and new findings or others. Hope this helps.

both are fine concerning your research originality. You either set up a new PCR-RFLP or if there is a method in the literatue that has been used successfully to screen for the same mutation in any other type of disease you can certainly use that. You will be the first to show that the method works for your particular disease theme for the first time ever, so there is your originality and actually the most important thing. Use an established one might be also a safer option since it has been proven before.
You can also try your new method and compare them.
So use eiher and i think you will be fine.

Either, but check wich options of endonucleases will cut/not cut your SNPs. You might find several options including isoschizomers that are cheaper or work better. Also worthed is to try the new enzymes that do the job in 15minutes.
