We had a discussion on whether the leukocyte population can shift between different samples on the FSC / SSC plot and should the gate be adjusted on individual samples therefore? We are working on humanized mouse model and leukocyte gate adjustment sometimes gives significantly different results on frequency of human CD45+ cells when it adjusted between samples or not.

Did anyone experienced anything similar during hu-mice model validation? Do you keep your leukocyte gate the same between samples once set?

I will appreciate if any other humanized mouse researchers can input on this. Thanks!
