Dear RG community,

We recently analyzed some extracts containing sulfated polysaccharides from different algaes and we wanted to test it for pyrogeineicity and endotoxin level using the LAL (Limulus amebocyte lysate) test. Although I have taken a lot of precaution during the extraction, including washing the algae extensively, working in sterile environment and purify them by ultrafiltration (MWCO=30kDa), I got results that are beyond the upper limit of the test. As to identify the issues, I was wondering if sulfated polysaccharides can interfere with the test ? And if yes in which extent ?

Are they solutions or alternatives to that test ?

Note: the extract contains various heteropolymer containing a wide range of sugar (arabinose, xylose, glucose, rhamnose, galactose, etc.) in various proportion, and sulfate groups.

Thanks for your help !

Arthur
