I am interested in measuring the decrease of catalase activity in samples of different organs (of mice), and groups (treated with different pollutants/heavy metals) kept in 4⁰C for a period of 40 days.
Does anyone know the effects on catalase activity when preserving samples at 4⁰C for short or long periods of time?
Catalase is a tetramer. Speaking generally, the hydrophobic interactions that hold the subunits together may be weakened at low temperature, especially if there is salt in the buffer. Also, 40 days is a very long time to keep the protein in solution. If it is not completely free of protease contamination, it could be degraded. Oxidation could also occur.
Good answer Mr Shapiro, the activity in the low temperature decreases significantly also 40 days is not possible to maintain the protein for this long period.
