The vial I have contains 2000 Kunitz units / 25mg of the enzyme. I want to make up the stock, but I need to decide what the working concentration should be. It would be used for a lung endothelial cell prep, in a 1h collagenase+dnase digest after physically mashing up the tissue.
I have used it for preparing neuronal cultures during 30 min digest. 2000 KU/mg 2 mg in 20 ml digestion solution.
Hi Troels, thanks, but this sounds like a lot more than I would expect, hmm. Actually, I meant to say that I have 0.25mg in my vial (not 25), sorry.

I use DNase in my preparation of tumour epithelial organoids following collagenase-trypsin digestion. I also have a vial of 2000 Kunitz unit / 0.25mg enzyme - to make my stock solution I dissolve this in 1 ml of PBS and make use of a 2U / ul working solution in order to break up organoid clusters and detach single cells.
