As you Know the pTZ57R/T vector in TA cloning procedure without insert can not close and so the gene of ampicillin resistance doesn't express. So why the vector without insert can growth on LB medium with ampicillin and produce blue colony?

Hi Ali,
It is not that surprising to get some background with homemade but also with commercial vectors. That is why you are advised to do blue/white screening. With the appropriate PCR product you will probably get much more colonies which will not turn blue.
