pDisplay has three restriction sites...BamH I, Bgl II and Pst I.

Check the restriction sites of your gene of interest on NEB cutter site to confirm that your gene start codon is in frame. Knock out your STOP codon with one of your restriction enzymes. Following heat shock transformation, (Growing up/ minipreps/ restriction digest to confirm band size) Check that your gene of interest is correctly orientated  with the Ig κ-chain leader sequence at the N-terminus and the myc/PDGFR-TM peptide at the C-terminus. Select the correcttly orientated constructs and do a endo free MAXIPREP so you have plenty of construct to repeat experiments with. Then transfect your cell line of interest and following the appropriate interval confirm expression  using a c-Myc antibody
