thank you very much for your suggestion. This cell line seems quite interesting even if it is not exactly what I was looking for (which are stem cells isolated from glioblastoma without any kind of stabilization/immortalization). I will contact the curator to see if I can buy these cells but I'll also continue to look for other suitable cell lines.
can you please be more precise? what kind of cell line are you referring to? Are these commercially available? Do I have to select them using the same procedure used for GSC isolation from solid tumors?
I don't know glioblastoma cell lines but it's possible from a cancer cell line to isolate few stem cells. I did extraction from prostate cancer cell lines or primary tissue. You can try to use the method used in these papers:
Pro-inflammatory gene expression in solid glioblastoma microenvironment and in hypoxic stem cells from human glioblastoma.
Tafani M, Di Vito M, Frati A, Pellegrini
Human prostate cancer initiating cells isolated directly from localized cancer do not form prostaspheres in primary culture.
Chen S, Principessa L, Isaacs JT
Cell-to-cell signaling influences the fate of prostate cancer stem cells and their potential to generate more aggressive tumors.
my search on this for breast cancer leaded me to using flow cytometry in particular (FACS)... i want to expose my cell to hypoxic condition to enhance its content of CD44 expressing cell....
I'd like to thank you all for your kind suggestions.
I actually know how to isolate these cells from tissues as we have both equipment and protocols. I won't have brain tumor tissue in the near future but I really need to confirm some molecular data we obtained in the past from these cell type (that we no longer have). A ready to use source is what I prefer. I'll try to contact the person suggested by Cristina.
