I would like to assess the editing efficiency of designed guide RNAs by transfection prior to establishing a stable KO cell line (via lentiviral infection). HEK293T cells are very efficient when assessing gRNA targeting human genes. However, I will be using a mouse model in parallel with the stable cell lines and would like a stable mouse cell line. Any suggested cell lines would be appreciated, along with the transfection media used and observed transfection efficiency.
Also NMuMG cell line is a breeze. But basically try using a cell type that is similar to your experimental design, or even better use the cell line you plan on using for the experiment but optimize your transfection protocols by using a EGFP vector and using Flow cytometry to quantify the transfection efficiency
Thank you so much! I will optimize the transfection conditons of Hepa1-6 which is another heptocyte cell line but is easier to transfect when compared to AML12 cells.
