I am studying novel immuno therapeutic strategies in cancer and have had trouble in establishing a reliable system to study combination therapy with PD-1 blockade. I am wondering if anyone has had success with such a model. I have tried B16 tumors and EL4 tumors. Thank you.

I suggest you to take a look to these two very recent papers. The anti PD-1 therapy has been performed using MC38 colon carcinoma and EMT6 breast cancer cells by Zippelius et al., CIR, 2015, and spontaneous mouse models of melanoma and breast cancer metastasis by Mittal et al, CR, 2014. Several years ago in an attempt to develop Multiple Myeloma tumors growing in SCID mice I grew a tumor. Further analysis revealed that it was an anaplastic sarcoma of mouse origin. The tumor was not human and it was not Myeloma, so I have no need for this tumor. It grows in SCID mice. I did not attempt to grow it in vitro but based on its extremely rapid growth in vivo, I think it would also grow in vitro. If you want it, feel free to contact me and I will provide it to you for your studies.
Xenograft model derived from B16 cell line is so useful that they differentiate (become to produce melanin) under the influence of CTLA-4 blockage. In the case of human malignant melanoma, not a few cases showed non-melanin melanoma and exhibit high metastatic potential. That is why the B6 WT mice transplanted of B16 with Matrigel is very useful.
EMT-6 models and MC38 models are excellent models for anti-PD1 and anti-PD-L1 treatment.
Recently, a co-worker Jesús Vera at Texas TU, finish succefulness a study using B16 tumor cell línea in C57BL/6 mice melanoma model to probe antiPD-L1 blockage alone por in combination with experimental treatment.
MC38, CT26, AT3 as subcutaneous tumors should respond to anti-PD1. The efficacy also depends on the anti-PD1 clone and the tumor size when you start the treatment. Usually you want your tumor to be around 20-25 mm2, 7-10 days after injection, then you can initiate your treatment.
I realise this is an old thread - I'm having trouble getting hold of anti-mouse PD-1 (clone G4 ideally), would any one have access to this (and be willing to ship to Belfast, expenses paid!) or recommend a commercial alternative?
