I am looking for an instrument that can measure supervisor's preferential treatment in workplace, which can be assessed by the responses obtained from the subordinates. There are several forms of preferential treatment and my focus is mostly on favouritism, nepotism and cronyism. It would be of great help if I could some help.
Leader Member Exchange theory (LMX) might be useful. It focuses on the quality of the relationship between the leader and the follower, distinguishing between followers who are in the leader's outgroup and in the leader's ingroup. The LMX 7 seven question instrument (Graen & Uhl-Bien, 1995) measures the quality of the relationship between the subordinate and the superior from the subordinate's point of view.

I agree with David, use of the LMX 7 will give you a good picture of the dyadic relationship between leader and member. But, this only refers to the relationships and preferences, not to actual differences in treatment.
Even so, that's the best tool I know of that could help you.

targeting employees "only" may be misleading. try to prepare a survey questionnaire of three categories.
1. For employers (or represented by executives)
2. Employees
3. Retired individuals
You may wish to randomise your study. The variations may help you eliminate bias. I would start like that.
