In our laboratory, we are stating research work on sirolimus production from natural isolates from soil and other sources. As per literature searching, high titre of sirolimus produced from Streptomyces hygroscopicus are documented, therefore we focus on this isolate from natural environment or other extreme environment? Anyone please guide me for starting basic work at lab scale.
soil samples can be dried and used for Streptomyces isolation. Then you can use species specific 16S r-RNA primers for screening the group you needed from the isolates.

Air-dried samples (as suggested by mr. Viswanath) contain a lower number of bacteria, reducing competition in isolation plates;
Semi-selective culture media may improve Streptomyces growth instead other genus. Think about addition of antibiotics as streptomycin even rapamycin and others to culture media. Perhaps in a population enrichment strategy.
Macro (colony) and micro-morphological characteristics may help to reduce the number of isolates candidates to molecular identification (think about using genes encoding key enzymes in sirolimus sinthesis pathway).
