I'm trying to find the promoter sequence of some cadherin family genes but cannot. If any of you know how to find out the promoter of a gene, please tell me.
Go to one if the genome browsers where you can filter for and export/download this info. Browsers are: UCSC, Ensembl and NCBI.
Try going to http://genome.ucsc.edu/ then click the link to Genome Browser. Type the name of your gene and you will zoom to its position in the genome. From there, browse the links to find and download whatever you need. You can set filters to show the number of base pairs of the promoter region you want upstream of the transcript start site. All gene features are annotated with hyperlinks to extract the relevant sequences.
Similar deal in Ensembl. Go to http://www.ensembl.org/index.html then select human genome, search for your gene name, then follow the links to locate the DNA sequence you need. You can export the data in multiple formats including FASTA, or just highlight the sequence and cut/paste into whatever software you use for downstream manipulation.
I agree with Heather. Web-based genomics can help you identify conserved promoter sequences.
Functional studies can also be very important. Cloning upstream regions into reporter gene assays can help you learn about expression control of your gene of interest.
