I need a easier and cheaper method than HPLC analysis like chemical analysis or spectroscopic method to determine the levels of glycosid steviols in Stevia rebaudiana extracts.
For qualitative and quantitative analysis of any herbal extract for phyto-chemical HPTLC is very dependable and faster technique than many others. For Steviol glycosides please refer..... 2 articles given below
1. Evaluation of steviol and its glycosides in< i> Stevia rebaudiana leaves and commercial sweetener by ultra-high-performance liquid chromatography-mass …

... was also proposed to evaluate steviol-glycosides [23], together with a validated HPTLC procedure
with ... detection [24] and a NIRS procedure for the quantification of steviol-glycosides [25 ... Recently,
2. a semi-quantitative determination of ST-glycosides was performed by desorption
Yes actually the HPLC method is most common and complete way to analysis and determine the stevia glycosides, but for some reasons we are not able to use the HPLC in lab, so i m going to find a replacement method to determine the levels of stevia glycosides in extracts
1. Understand the research papers on HPTLC given above for HPTLC of "steviol glycosides in stevia rebaudiana extracts"
2. Try to develop small TLC plates (Silicagel 254 G.) and preserve the developed plates for scanning under CAMAG HPTLC scanner. (There must be one or two R&D Lab in Iran with HPTLC scanners and request them for some cooperation to scan your plates)
3.You may contact CAMAG distributor in IRAN ( as given in their sites.... M/s MAHZAD KALA CO. LTD.
if you want to quantify glycosides, I suggest HPLC, with an optimal extraction of the leaves, however if you want only to detect glycosides, you can use thin layer chromatography TLC.
