I am interested to measure oxidative stress (MDA, 4HNE, 8OHdG) and antioxidant (SOD, GPx, Catalase) in SD rat sperm. Does anyone have protocol or reference or expertise to measure these parameters? I really appreciate your cooperation.
See our works, maybe you find something interesting. We could measure 8-oxodG (and other DNA modifications) commercially. Please contact directly if you are interested.

Daniel

Hello! For the MDA determination (Free MDA and Protein MDA) our lab has developed and published a method. See the link below for the abstract. It is a very credible method and works fine with any kind of sample.

You can find attached below the full article. As you will read in the «Sample treatment» section, you can use any kind of sample. You can use a frozen sperm sample but you need to let your sample thawn to RT first. After that, you must isolate the protein fraction of your sample (you can find the detailed process of handling your sample in order to isolate the protein fraction in our paper in the «sample treatment section»). Then you can measure the Protein MDA in the protein fraction, as described.
Check our paper and let me know if you have any further questions!
There are plenty of good protocols. We recently described a simple and rapid method for measurement of total reductive (antioxidant) capacity with very good precision and accuracy:
