Some studies use fluorescent-labelled cells for transplantation in knee or in any bone for tracing the fate of transplanted cells:
I really want to know if the fluorescence is lost due to decalcification or not? If yes, what decalcified method was used. If not, were plastic sections used,
For example some have used Dil. Please provide me a working procedure if I inject a fluorescent-labelled cells into knee and I can trace them. My main concern is decalcification.

in general, acid decalcification will not help too much. Formic acid and its combinations will do much harm and results in autofluorescence. EDTA should be tried, and in general is OK, but when you want to visualize you sample, make sure the pH is compatible with fluorescence for your dye/label. For many dyes, fluorescence and fluorescence life time is pH dependent and can result in no signal experience if the conditions were not well set.
