We are trying to transfect these cell lines using Lipofectamine 3000. It works with GFP, but it doesn't work with a larger construct. Do you have any suggestions? We are currently following the protocol and adding 5 ug DNA/well in a 6well plate to complete media (with FBS and P/S) and leaving it on for 6hours.

Hi, my suggestion would be to know the efficiency of transfection, doing a titration of 2, 4, 6, 8 ug DNA/well. Exposure time may also needs to be optimized. You may increase the exposure time from 6 to 8,10,12 hours. Best wishes.
