You can try primers Mtb1 (5'-CCG-GCG-GGG-CCG-GCG-G) and Mtb2 (5'-CGG-CGG-CAA-CGG-CGG-C)

Please follow any publication with nested PCR for TB diagnosis, so that your PCR could be highly specific too, to avoid false positives.

I want to detect NTM from drinking water, I need to design primer for each species or I can use universal primer for 16S rRNA sequencing. how can I design primer for each species of NTM?  Also for culturing how can I recognize colonies for NTM? kindly advise me
