We are trying to retrofit a cryogenic cell phone grinder for use in grinding large amounts of ovine brain and liver. We are planning to spray CO2 from a tank containing liquid CO2 onto the samples. We will probably have to pre-freeze the samples with dry ice. Most researchers use liquid nitrogen even for small samples. It is possible that we will have to cut the samples up into smaller sizes. Any experience would be helpful.
Kenneth: How did you freeze the horse spleen/liver? Did you use liquid nitrogen or perhaps dry ice? I am having some difficulty finding out what temperature the tissue needs to be at before it reaches this brittle stage. After breaking them into smaller pieces did you refreeze them or did you immediately place them in the grinder? What kind of tissue grinder was this? The one we are using has a cylindrical cutter of about 2" in diameter. When the grinder is turned on, this cylinder rotates at a high speed. The tissue would be forced onto the grinder with a plunger. Remember, this was initially used to grind cell phones. Thanks for your reply.
