I want to elicit PTI responses in mesophyl protoplasts using chitosan. I have tried treating them with 20ug/ml (1/5 of what is reported in the literature for intact leaves) but this treatment appears to lyse them as there is no pellet observed after treatment compared with the control. Has anyone seen this before and what do you think might be happening? I don't think chitosan would trigger HR.
