We have a Leica DM IL inverted microscope. Can anyone help me out on how to switch to the fluorescence settings? When viewing my stained slides.. Instead of a black background, I am viewing a grey one. Please help.
Hi Jhoti, pay attention to switching off transmitted light when you use fluorescence and also ensure that no room light illuminate directly your sample (best would be a dark room). If you have a problem deriving from light coming from the environment an option is just preventing light hitting your sample using some cover (even a small cardboard box).
knowing, this my answer is coming really late (unfortunately I found the question some day ago)... Besides the hints Fabio gave in his reply above, I would suggest the following:
Normally a Leica microscope always comes with a manual of operation. I guess you have the "old version of the DM-IL" (with inbuilt glow-lamp). But it could be also that you have the new version DM-IL-LED. For both versions there are several documents to be downloaded cf:
Have you followed the instructions of the manufacturer already? If yes, then tell us -please- more about your troubles (do you have installed the additional incident light fluorescence unit,? lamp, fluorescence slide for the three filter cubes, etc.).
If you can't see fluorescence on a dark (black) background, adhere to Fabio's tecnical tipp/advice (cardboard cube or cylinder) between object slide and ligth source as well as below the slide platform.
But I guess you may have overcome the problem earlier than our answers were given.
