Hi guys, recently I collected few clinical samples for FISH and histology in methacarn and 10% NBF respectively on blotting paper to maintain orientation. But during the step of removal of fixative and change it with 100 % alcohol, the tissue got detach and is now freely floating in the eppendorf. Can someone please devise some method to orient it? so that I can give samples for paraffin embedding?
Hello, unfortunately there is usually no chance to orientate oneself macroscopically on intestinal biopsies. At slightly larger mucosal biopsies it may be possible to orientate oneself by means of the musccularis mucosae (even if it is not always clear on which side the mucosa or submucosa is). I have never had macroscopic orientation on biopsies of the tunica muscularis.
sorry, I guess I missed your question with my previous answer. You could use patholigical ink (or anything like this) for maintaining orientation and fix the samples in formalin. This should work for paraffin-embedding.
Please find an attachment, where it is suggested that endoscopic biopsies can be oriented after fixation.
Since the tissue is very small(0.5 mm), inking step might ink the entire tissue, correct me if i am wrong.....
