I am trying to examine the vascular permeability of certain compound. I think this compound may have an effect on vascular permeability. I am working on porcine isolated coronary arteries. My question is about any recommendation for certain methods to investigate the vascular permeability.

There's several ways to assess vascular leak.
One classic method is use of Evans Blue dye, which binds to albumin. Healthy vasculature is impermeable to albumin, while tissues with leaky vasculature will accumulate blue dye. This can be then extracted and assessed with a spectrophotometer.
An alternative is to use labeled dextran or another macromolecule. The concept remains the same.
Normally, these are used in vivo, but if you have just excised vessels you could probably use an ex vivo perfusion system.
