Hi!
I want to transfect murine hematopoietic progenitors with a transposon plasmid.
We have a Lonza nucleofector and it seems that this is the best way to get DNA in those cells. We have tried DK-100 and DN-107 programs, however, the post-electroporation viability of hematopoietic progenitor cells decrease significantly.
Therefore, I was wondering if you have any suggestions regarding nucleofector protocols for such cells.

Check the nucleofection buffer and the recommended cell densities and volumes. For human HSPCs the Lonza programs works well.

Thank you, Hasan Iss. We have checked the nucleofection buffer and the recommended cell densities and volume for mouse HSPCs , but the result is still not very good.
