you can easily measure stomata opening and closing by (Infra Red Gas Analyzer) IRGA !!

Please have a look on the following publications which used indirect method for finding stomatal opening and closure (PSII efficiency under non-photorespiratory conditions). If you used this method it give you also a whole picture for stomatal status of whole leaf
