I have to do tobacco transformation using pGreenII 0229 binary vector system. Does it works the same as that of any other binary vector in tobacco? can anyone provide me any advice or suggestion regarding this? Is there anything I should now about before starting the experiment? Can anybody provide me an reference article about it?
Doesn't pGreen plasmid series need to go with another plasmid for transformation!?

In my previous lab, one student tried to perform Agro transformation of potato using above mentioned vector, soon he had to change vector as he was not getting expected regeneration response. May be what Prof. Yuan said stands true here. I think he was using EHA101 strain.
The attached paper showed that pGreen0229 was co-transformed with plasmid pSoup to work (see yellow highlights on p.4178).

I already have transformed pGreenII 0229 (containing gene of interest), along with pSOUP in Agrobacterium strain EHA105. I was just wondering if anyone has came across any articles mentioning 'tobacco transformation with pGreen II 0229 vector'. Thank you Yuan-Yeu Yau sir for the for the article.
I am not sure whether pGreen0299 is the same as pGreenII0299 (as you mentioned). But pGreen0299 had been used for genetic transformation of tobacco. See the attached paper. I am very curious why they did not mention 'pSoup' in the paper.
