It looks like there is another thread explaining similar difficulties with BT549 in even more immunocomprimised mice. Any chance you can choose a differrent cell line?
Since the beginning of this year, I also tried this model. I first tried Nude-Foxn1nu mice (5 M cells with matrigel), but no tumor developed.
About 2 months ago, I tried to inject the same number of cells in B6;129-Rag2tm1FwaII2rgtm1Rsky and NOD.CB17-Prkdc^scid mice and there were no tumor growth in the past 6 weeks. Until this morning, I suddenly reallized that 2 Rag2tm1FwaII2rgtm1Rsky mice among 5 developed tumors about 300 mm3 now. However, no NOD.CB17-Prkdc^scid mice develop the tumor yet.
There are several reports showed that BT549 growth in vivo, but I just feel this cell line is really hard to grow in vivo.
Thanks Ning for the information. RAG2/IL2RG mice are more immunodeficient than NOD SCID mice-that's why you saw tumor development. I guess increasing cell number to 1x107 per injection might increase the tumor take? Also, NSG mice or NOG mice might yield similar or better results.. Just to make sure, you injected 5 million cells in Matrigel subcutanously?
Yes, I injected 5M cells with Matrigel 1:1 ratio. And the injection site is the 4th mammary pad (without operation, just injection).
However, I am little bit doubt about the injection of 10M cells because BT549 cells are bigger than most of the BC cell lines and I just feel it hard to prepare such high density of the cells especially with Matrigel.
