I have trouble with exciting wavelengths higher than 460 nm while e.g. ex with 405 nm works. Filtercubes are correc, as I used them with my old xenon lamp before... I work on axiovert200M and with slidebook. And new with coolLED pe 4000 (liquid lightcable).
Check with your local rep first to make sure that everything is working properly. If you wanted to fix things on your own (make sure that it does not void the warranty!):
- Are you seeing the light coming out of the objectives with different filter settings?
- Seems there are a lot of settings with the pE-4000 series as I see from the user manual (https://www.coolled.com/wp-content/uploads/2018/07/pE-4000-User-Manual-DOC-008-Iss-6.pdf). Check if the controls are set to the right levels for each wavelength that you are interested.
- Best course is to contact your local rep and making sure if everything is fine
There can be anything from simple setup to something blocking the light on its path, to a faulty system, etc. etc. Until you work with the system in person, you cannot provide an answer of what is happening.

Thank you Sathya Srinivasan . Yes, that's the way I noticed the problem: As I have been using a monochromator as light source before, my filter cubes had no excitation filter. And the new light source has much more power, but the excitation light is not that small in its bandwidth as I expected from these LEDs (I did an excitation with 460 nm and it shined through till about 500 nm and therefore through my dichroic and my emission filter!). Same for higher wavelengths, too. I saw it by the way you wrote, too: the light is coming through the objectives!
So, I have to add now additional excitation filters (which can be easily applied directly at the light source, to enable quick changes....but additional costs for brackets and filters I didn't expect.. :( )
